Composition
Each tablet contains:
- Tamoxifen Citrate – 10 mg
- Excipients – q.s.
Pharmacological Effects
- Selective Estrogen Receptor Modulator (SERM):
- Antiestrogenic (in breast tissue): Blocks estrogen receptors, inhibiting estrogen-dependent cancer growth.
- Estrogenic (in bone/liver): Preserves bone density & improves lipid profile.
- Partial Agonist/Antagonist: Effects vary by tissue type.
Indications
- Breast Cancer Treatment & Prevention: Adjuvant therapy for ER+ breast cancer (women & men).
- Risk Reduction: In high-risk women (e.g., BRCA mutation carriers).
- Infertility (off-label): Used to stimulate ovulation in PCOS.
- Gynecomastia Prevention (off-label): During androgen therapy.
Recommended Dose
- Breast Cancer (Adjuvant): 20 mg/day (10 mg BID or single dose).
- Prevention (High-Risk Women): 20 mg/day for 5 years.
- Gynecomastia Prevention: 10–20 mg/day (often cycled).
- Infertility (PCOS): 10–20 mg/day (days 3–7 of cycle).
Side Effects
Common: Hot flashes, vaginal dryness, menstrual irregularities.
Serious but Rare:
- Endometrial cancer (increased risk with long-term use).
- Thromboembolism (DVT, pulmonary embolism).
- Ocular changes (cataracts, retinopathy).
- Men: Reduced libido, erectile dysfunction (rare).
Contraindications
- Pregnancy: Category D – risk of fetal harm.
- History of Thromboembolism: Contraindicated in patients with prior events.
- Concurrent Use: With aromatase inhibitors (no added benefit).
- Hypersensitivity: To tamoxifen or formulation components.
